Hydrated cartilage maintains its compressive strength and elasticity, acting as a biological shock absorber. Without adequate chondroitin, cartilage progressively loses water content, becoming brittle and less able to absorb the mechanical loads placed on joints during everyday movement.

Evidence: Multiple meta-analyses of randomised controlled trials have found statistically significant improvements in pain and function with Chondroitin Sulfate supplementation. Boswellia Extract — Joint Brex Ingredient 3

Role: 5-LOX enzyme inhibitor. Boswellia serrata resin contains boswellic acids — triterpenoids that specifically inhibit 5-lipoxygenase (5-LOX) enzymes. These enzymes drive leukotriene production, a primary mediator of chronic joint inflammation. By inhibiting 5-LOX, Boswellia reduces leukotriene-driven inflammation without disrupting the prostaglandin pathway — which is why it does not carry the gastrointestinal side effects associated with NSAIDs.

Role: NF-kB signalling reduction. Multiple systematic reviews have found consistent pain reduction and functional improvement across populations with joint conditions. Limitation: Plain curcumin has limited oral bioavailability. Formulations with piperine (black pepper extract) or phospholipid complexes absorb substantially better. MSM performs best within a combination formula rather than in isolation — its connective tissue support complements the cartilage-rebuilding action of Glucosamine and Chondroitin.

Limitation: Effect sizes in isolation are moderate. Its primary value in Joint Brex is as part of the connective tissue support layer within a multi-ingredient formula.

Bromelain — Joint Brex Ingredient 6

Role: Proteolytic enzyme for inflammatory protein clearance. Bromelain is a cysteine protease enzyme extracted from pineapple stem. Unlike small-molecule anti-inflammatories, it works enzymatically — physically cleaving inflammatory protein complexes (fibrin, immune complexes, kinins) that accumulate in swollen joint tissue and restrict circulation.
